Is there a way to associate Comboblocks Readmore links with the page or post titles they link to for accessibility sake?

Welcome to PickPlugins.
I'm not sure if you're using a shortcode-based post grid or a Gutenberg block, but the feature is available for both.
For shortcode-based grids, you can expand the "Read More" layout element and choose your preferred "Link To" option. Here's a screenshot - https://ibb.co/p11J6V8
